PAF Acetylhydrolase Assay Kit Reference: 760901-96 PAF-AH catalyzes the hydrolysis of the potent biologically-active phospholipid PAF, generating inactive lyso-PAF. Cayman’s PAF-AH assay kit provides an accurate and convenient method for measurement of PAF-AH activity. The assay uses 2-thio PAF which serves as a substrate for PAF-AH. Upon hydrolysis of the acetyl thioester bond at the sn-2 position by PAF-AH, free thiols are detected using Ellman’s reagent.
sPLA2 Assay Kit Reference: 765001-96 Cayman’s sPLA2 Assay provides an accurate and convenient method for measurement of sPLA2 activity. This assay uses the 1,2-dithio analog of diheptanoyl phosphatidylcholine which serves as a substrate for most PLA2s with the exception of cPLA2. Upon hydrolysis of the thioester bond at the sn-2 position by PLA2, free thiols are detected using DTNB.
cPLA2 Assay Kit Reference: 765021-96 Arachidonoyl thio-PC is a substrate for cPLA2 by virtue of the presence of arachidonic acid at the sn-2 position of the glycerophospholipid. Hydrolysis of the arachidonoyl thioester bond at the sn-2 position by PLA2 releases free thiol which can be detected by DTNB. This assay can be used to determine the activity of cPLA2 in purified preparations, cell cultures, or tissue homogenates that are known to contain only cPLA2. Use of this assay with preparations containing more than one type of PLA2 will result in the measurement of total PLA2 activity rather than cPLA2 alone. Isozyme-specific cPLA2 activity can be measured by excluding sPLA2 or inhibiting iPLA2 activities in the assay.
Nitrate/Nitrite Colorimetric Assay Kit Reference: 780001-2 Cayman’s Nitrate/Nitrite Assay provides an accurate and convenient method for measurement of total nitrate/nitrite concentrations. This kit can be used to measure nitrate and nitrite in plasma/serum, urine, tissue culture media, and tissue homogenates. However, it cannot be used to analyze nitrate and nitrite from an in vitro assay of NOS in which excess NADPH has been added. For these assays a second method (LDH method) is utilized and is available from Cayman in a 96-well plate format (Item No. 760871).
Nitrate/Nitrite Fluorometric Assay Kit Reference: 780051-2 Cayman’s Nitrate/Nitrite Fluorometric Assay provides a convenient method for the quantitation of low levels of nitrate and nitrite in biological samples (particularly tissue culture medium). Previously undetectable concentrations of nitrate/nitrite in a variety of samples can be accurately quantitated using this kit as it extends the lower limit of detection by 20-fold over our colorimetric version (Item No. 780001). The minimum detectable quantity of NO2/NO3 is ~50 nM.
NOS Activity Assay Kit Reference: 781001-1 The NOS Activity Assay measures NOS activity by monitoring the conversion of radiolabeled arginine to citrulline. This assay is simple, sensitive, and specific for NOS activity and can be used with both crude and purified enzyme preparations. The kit includes sufficient materials and reagents for 50 total reactions. Radiolabeled arginine and NADPH are not included with the kit.
Caspase-8, 10 Activity Fluorometric Assay Kit Reference: RP10238K Caspase-8, 10 Activity Fluorometric Assay Kit can be used for assaying caspase-8, 10, 3, 7 activities in cell/tissue extracts in a 96-well plate format. Each of the supplied fluorogenic substrate and the pan caspase inhibitor Z-VAD-FMK are sufficient for 250 x 100 μl reactions. The supplied TRAIL-treated cell lysate is used as a positive control, sufficient for 5 X 100 μl reactions.
